Subject: Post-doctoral researcher in Organic Chemistry: Molecular engineering of peptidic polymers to coat Gold NanoRods for biomedical applications

In the frame of an ANR project (SmartNanoCoat) focused on the rational design of new structurally controlled polyfunctional multidentate polymers to coat inorganic nanoparticles in view of their biomedical applications, a 12-month post-doctoral position (renewable 6 months) in chemistry is available at ICSN. More precisely, the candidate will integrate a group composed of 3 permanent researchers in the Chemical Biology department (ICSN). For the nanoparticle aspects, the work will be realized notably at Institut de Recherche de Chimie Paris (IRCP).

In the context of the research in innovative and efficient systems for both treating and detecting diseases, NanoParticles (NPs) offer modular in vivo pharmacokinetic and targeted biodistribution in diseased tissues. However, while many inorganic NPs have been identified as promising since the 90’s, less than 10 are nowadays approved for patient healthcare.
Toxicity issues related to colloidal instability in biological media and functionalization control/characterization difficulties are important unsolved drawbacks that remain serious bottlenecks in the development of inorganic NPs for biomedical applications.
To overcome these issues, we propose the development of new multidentate polymers with a perfectly defined structure to coat NPs in a highly stable and stealth manner. In this project, we will explore an innovative approach to design and synthesize multidentate functionalized peptides, before validating their potential to coat Gold NanoRods (GNRs) for biomedical purposes. This strategy is modular and tunable, allowing a rational design of inorganic NPs coating depending on the desired properties of the final NPs. The rod-like gold NPs are especially attractive, due to their unique optical properties and potential applications in functional nanodevices thanks to photoacoustic, radio-enhancer or photothermal properties. This project, going from building block synthesis to preliminary in vivo evaluations will pave the way to the development of GNRs as targeted nanomedicines (e.g. tumor photothermal ablation).

The selected candidate will be in charge of the synthesis (and characterization) of:
- the amino acid derivatives (RMN/LC-MS characterizations)
- the multi-anchoring peptide ligands with solid phase synthesis (LC-MS/Maldi characterizations)
- the Gold NanoRods by seed/growth preparation (UV-Vis/TEM characterizations)
She/he will prepare (and characterize) and evaluate/optimize the new ligands on Gold NanoRods (UV-Vis/TEM/XPS/QCM/AFM characterizations) in biological media and on cells.
She/he will be in charge of lab-book/reports/publications redaction.
The post-doctoral fellow will need a strong background in organic chemistry but will have also to adapt her/himself to the multidisciplinary of the project.

The prospective candidate will have a Ph.D. in Organic Chemistry and proven research skills evidenced by at least one publication as the first/joint first or second author.
We are looking for applicants with a strong background in organic synthesis, with a good knowledge in peptide synthesis, willing to work at the interface between Chemistry and Material Science for biomedical applications.
Interdisciplinary profile, especially peptide, nanoparticle and surface chemistries will be appreciated.
Candidates must have organisational abilities and a very positive attitude to working in a team on multi-disciplinary projects while respecting rules and procedures.
She/he should be fluent in English (and ideally also in French).

Context of work:
Gathering a 200-collaborator staff, the ICSN is the chemistry Institute of the CNRS campus in Gif sur Yvette (south suburb of Paris). The Institute belongs to the Paris-Saclay University Campus, which will incorporate 10% of the French research. The ICSN develops research activities at the interface between chemistry and biology, that draw inspiration from natural substances. The Chemical Biology department (link) is one of the four departments of the institute that beneficiate of an exceptional working environment for chemistry/biology with wide service facilities (link).
The IRCP (link) is a mixed research unit of the CNRS and Chimie ParisTech (French High-School). The Institut highlight an integrated research that cover a wide range of fields of chemistry: from molecular to materials chemistry. It beneficiates of all facilities for chemistry, especially for nanoparticle evaluation/characterisation (XPS, QCM, TEM, AFM, ICP-MS…).

Constraints and risks:

Project activities will be share out between mainly two laboratories with periods of organic synthesis at ICSN and nanoparticles preparation/evaluation at IRCP that will involve a geographic mobility between these two labs (45 min by train).
